This is something that has no validity. All the graves of the Prophets are unknown, except for the grave of our Prophet Muhammad (peace and blessings be upon him) in Madinah, and the grave of Ibrahim in al-Khalil. Everything else is not known to the scholars of knowledge, and one should not believe whoever says otherwise, whether it is the grave of Ayyub or anyone other than Ayyub (peace and blessings be upon him).
Then, even if we assume that the grave of Ayyub or anyone other than Ayyub, or Imran or anyone other than Imran, exists, it is not permissible for anyone to worship it besides Allah, nor to call upon it besides Allah. Even the Messenger Muhammad (peace and blessings be upon him), who is the best of creation, it is not permissible for anyone to call upon him besides Allah, or to seek his aid, or to make vows to him, or to slaughter for him to draw near to him with sacrifices; because this is major shirk (associating partners with Allah). وَلا تَدْعُ مِنْ دُونِ اللَّهِ مَا لا يَنْفَعُكَ وَلا يَضُرُّكَ فَإِنْ فَعَلْتَ فَإِنَّكَ إِذًا مِنَ الظَّالِمِينَThat is Allah, your Lord; to Him belongs sovereignty. And those whom you invoke other than Him do not possess [as much as] the membrane of a date seed. (14) If you invoke them, they do not hear your supplication; and if they heard, they would not respond to you. And on the Day of Resurrection they will deny your association. And none can inform you like [one] Acquainted [with all matters].
